Biomarkers of behavior, therapeutic targets for adult B-acute lymphoblastic leukemia identified

New insight into the aggressive behavior of certain adult B-acute lymphoblastic leukemias has provided researchers with a potential new prognostic biomarker and a promising new therapeutic target.

The research, conducted by Ari Melnick, M.D., associate professor of medicine and director of the Raymond and Beverly Sackler Center for Biomedical and Physical Sciences at Weill Cornell Medical College and a hematologist-oncologist at New York-Presbyterian Hospital/Weill Cornell Medical Center, and colleagues, was published in Cancer Discovery, a journal of the American Association for Cancer Research.

Although B-acute lymphoblastic leukemia is highly curable in children, the disease is usually fatal in adults, and researchers have yet to identify why this is the case. Part of the explanation for the poorer outcomes in adults is the higher frequency of genetic alterations associated with unfavorable prognosis.

In order to better understand why these genetic alterations are associated with poor outcomes, Melnick and colleagues studied 215 diagnostic specimens obtained from adults with B-acute lymphoblastic leukemia who were participating in a large Eastern Cooperative Oncology Group phase III clinical trial.

"We performed an integrative epigenomics study to decode the instructions that determine how these cells behave," Melnick said. "The hope was that this would allow us to identify better survival biomarkers and new therapeutic targets."

In many cancers, genetic alterations work in conjunction with epigenetic changes (changes in the way that DNA is modified and packaged) to promote cancerous behaviors. Looking at the B-acute lymphoblastic leukemia specimens, Melnick and colleagues found that many of the leukemias' bad traits were a result of changes in the epigenetic code. In many cases, the epigenetic changes were directly linked to the proteins generated from the genetic alterations and could be used to identify key master regulators required for the leukemic cells to live, according to Melnick.

"For example, we found that a cell surface molecule called CD25 was an extremely powerful indicator of the presence of the most aggressive and fatal cases," Melnick said.

The researchers also discovered that abnormal forms of the E2A and MLL proteins occurring in B-acute lymphoblastic leukemias directly reprogram epigenetic settings at their binding sites throughout the genome.

Most notably, the researchers found that mutant forms of MLL epigenetically reprogramed leukemia cells to express the powerful oncoprotein BCL6, and that BCL6 was required to maintain the proliferation and survival of the leukemia cells.

"We then designed inhibitors of BCL6 and showed that we could kill leukemia cells from patients enrolled in the clinical trial by blocking its function," Melnick said.

Based on these results, the researchers plan to use CD25 as a biomarker to identify those patients who have the worst disease in the next set of clinical trials, and to tailor treatment appropriately. In addition, BCL6 inhibitors are currently being translated for use in humans, and they hope to develop clinical trials targeting BCL6 in MLL-rearranged leukemias.

"These results will ultimately lead to biomarkers that help guide treatment and to the development of therapies that will be more effective for patients with this aggressive form of leukemia," Melnick said.

Hurricane Sandy: Economic Impact Could Exceed ... - Yahoo! Finance

Hurricane Sandy moved closer to the New Jersey coastline Monday morning and was expected to pummel shore communities with 85 miles per hour wind, extreme tides and severe flooding later this afternoon.

The National Hurricane Center has warned nearly half a dozen states including New York, Virginia, Connecticut and Pennsylvania to prepare for what many are calling a "Frankenstorm" ? a frightening mix of rain, snow and wind that's part hurricane, part nor'easter.

More than 60 million Americans are likely to be impacted by Sandy and experts are already forecasting damages in the range of $2 billion to $100 billion.

Hundreds of schools, offices, shopping malls and restaurants have closed their doors in anticipation of the hurricane. Broadway theaters canceled their shows. Atlantic City casinos sent gamblers home.

New York City suspended all subway and bus service and New Jersey shut down its public transportation. U.S. stock markets also halted both floor and electronic trading on Monday. The last time the New York Stock Exchange closed because of a weather-related event was on Sept. 27, 1985, when Hurricane Gloria pounded the East Coast.

Airlines have grounded thousands of flights in the Northeast; according to the flight-tracking service FlightAware, nearly 7,500 flights had been canceled for Monday and Tuesday. President Obama declared New Jersey, Rhode Island, New York, Pennsylvania and the District of Columbia as state of emergencies.

Ryan Sweet, a senior economist at Moody's Analytics, says Sandy would be a costly storm but one that would have little impact on fourth quarter gross domestic product.

"It's much more a regional story?[Sandy] will not have significant U.S. macro economic implications," he says in a phone interview. Sweet estimates the lost economic output between Washington, D.C. and New York City would equal at least $10 billion a day.

Diane Swonk, chief economist at Mesirow Financial, says Sandy could boost spending "fairly dramatically" as people stock up on food, water, batteries and other essentials before the storm hits landfall.

"This will show up in increased spending at hardware and home stores," she writes in a recent note. "There should also be an increase in spending, once damages from the storm are assessed and repairs get underway. That spending could borrow a bit from traditional holiday sales, depending on how much insurance is paid on those claims."

The closure of supermarkets, retail stores and restaurants could "negatively impact retail sales by half a percent," according to Lance Roberts, CEO of Streettalk Advisors. Sandy's damage may result in lost wages and a rise in unemployment claims in the short term, he says, but hurricanes can also boost GDP depending on the severity of the damage and cost to homeowners and local businesses.

Sandy's wrath could push up gasoline prices if oil refineries and oil ports in Philadelphia, Northern New Jersey and Delaware go offline, notes LPL Financial economist John Canally. Power outages are the most serious threat to businesses and consumers, he adds, but Sandy could have the same effect on retail spending as last October's abnormal East Coast winter storm: not much.

"People are sitting in their homes and not spending," Canally says. But "it's not like a week before Christmas" or another major holiday, he adds.

Hurricane Irene in 2011 cost an estimated $15 billion to $20 billion in damages.

Hurricane forces Obama to balance governing, campaigning

WASHINGTON (Reuters) - President Barack Obama faced the delicate task on Sunday of balancing his response to a potentially huge natural disaster with his own tough re-election effort as Hurricane Sandy bore down on the U.S. East Coast nine days before Election Day.

Trying to demonstrate that he had learned the lessons of White House predecessor George W. Bush's botched handling of Hurricane Katrina in 2005, Obama sought to project the image of a president fully engaged in marshaling resources to deal with a looming national emergency.

Visiting the federal storm-response headquarters in Washington, Obama warned East Coast residents to prepare for a "serious and big" storm that will be slow-moving and might take time to clear up. But he vowed the government would "respond big and respond fast" after it strikes.

Even as he pledged to stay on top of the storm threat, Obama - after shuffling his campaign travel because of the approaching hurricane - stuck to his plan to fly to Florida on Sunday night for a rally in Orlando on Monday.

But he scrapped an appearance later in the day in Ohio - considered the most critical election swing state - so that he could return to Washington to monitor what could be one of the largest storms to ever hit the U.S. mainland.

The hurricane threat also scrambled Republican challenger Mitt Romney's schedule, but he too was going ahead with some of his events.

With Sandy forecast to barrel ashore between the mid-Atlantic states to New England late on Monday, Obama faces an increased risk to his prospects in a tight presidential race if the government's emergency apparatus fails to perform well. Voters go to the polls nationwide on November 6.

Obama, who also held a conference call with governors and mayors from states in the storm's path, sought to allay such concerns.

"My main message to everybody involved is that we have to take this seriously," Obama told reporters after he was briefed. He said emergency officials were confident that enough equipment and personnel were in place in advance of the storm's onslaught.

"My message to the governors, as well as to the mayors, is anything they need, we will be there, and we will cut through red tape. We are not going to get bogged down with a lot of rules," he said.

LESSONS LEARNED FROM KATRINA

Obama's aides said storm preparations were in line with his response to other natural disasters on his watch, such as last summer's devastating wildfires in Colorado, last year's Hurricane Irene and the tropical storm that hit the Gulf Coast this summer ahead of the Democratic convention.

President George W. Bush suffered serious political damage from his administration's inept handling of Katrina, which devastated New Orleans early in his second term. He was also widely criticized for being out-of-touch personally and slow to respond to the crisis.

With that in mind, Obama's White House has made it a practice of trying to get ahead of events while putting the president front and center.

This time, however, the country faces a major natural disaster on the cusp of a presidential election, when the incumbent has recently been struggling to curb his opponent's newfound momentum.

While the risk of a fumbled government response could have a political cost, it is also an opportunity for Obama to look presidential in a national crisis in a way that Romney can't.

The pressure is on Craig Fugate, head of the Federal Emergency Management Agency, to prevent a repeat of Katrina.

FEMA bore the brunt of the blame for the Katrina debacle and then-director Michael Brown resigned shortly afterward, with Bush's remark "Brownie, you're doing a heck of a job" an enduring source of ridicule.

With Obama looking on, Fugate - who was appointed by the Democratic president - exhorted his lieutenants not to let down their guard.

Officials in the path of the storm scrambled to ensure that extended power outages would not disrupt early voting that appears to be critical for both candidates this year.

Obama said he did not think the storm would impact voting, but some on his staff were not so certain. A severe disruption is likely to hurt Obama more, as his campaign has counted on early voting to lock up the support of younger voters and others considered less likely to go to the polls on Election Day.

But an Obama campaign official said, "We remain confident in our ability to get our voters to the polls by Election Day."

Mindful of the hardships ahead, the Obama campaign said it would suspend fundraising emails in Virginia, Pennsylvania, North Carolina, New Jersey and Washington D.C., starting on Monday and encouraged supporters to donate to the Red Cross.

Gigantic Hurricane Sandy bears down East Coast

NEW YORK CITY (Reuters) - Tens of millions of people along the U.S. East Coast girded themselves on Sunday for Hurricane Sandy, a gigantic storm forecast to assault the densely populated region with battering winds, dangerous flooding and even heavy snowfall.

Sandy, expected to come ashore late on Monday, could deliver a harsh blow to major cities in its target zone including New York, Philadelphia, Washington, Baltimore and Boston. Its center was forecast to strike New York-New Jersey area and then move inland toward Philadelphia and the rest of Pennsylvania.

The sheer size of the storm meant its effects would be felt from the mid-Atlantic states to New England. Officials warned of widespread power outages that could last for days.

In New York City, subway, bus and train service will be suspended on Sunday evening and up to 375,000 people were ordered to evacuate from low-lying areas.

President Barack Obama, speaking after a briefing at the federal government's storm response center in Washington, called Sandy a "serious and big storm" and asked residents to heed the orders of state and local authorities to protect themselves from its onslaught.

"It's a very, very large system," National Hurricane Center Director Rick Knabb said. "The storm is going to carve a pretty large swath of bad weather, both water and wind."

The storm could bring the country's financial nerve center to a standstill, although the major Wall Street exchanges said they planned to open as usual on Monday because they have alternative facilities they can use.

Worried residents in the hurricane's path packed stores, searching for generators, flashlights, batteries, food and other supplies in anticipation of power outages.

New York City schools will be shut on Monday. Other local governments also announced school closures.

Gale-force winds were already starting to buffet Virginia and could reach other parts of the mid-Atlantic coast on Sunday night. Tens of millions of people will feel its bluster for as long as two days, Knabb said.

Forecasters said Sandy was a rare, hybrid "super storm" created by an Arctic jet stream wrapping itself around a tropical storm, possibly causing up to 12 inches of rain in some areas, as well as up to 2 feet of snowfall in the Appalachian Mountains from West Virginia to Kentucky.

It could be the largest storm to hit the United States, according to the National Oceanic and Atmospheric Administration's (NOAA) website.

Sandy was already disrupting transportation systems. More than 700 flights, including international ones, were canceled on Sunday and nearly 2,500 more were canceled for Monday, FlightAware.com said.

CASINOS CLOSE

New Jersey casinos were ordered to close and state officials decided to shut bus and rail systems by early on Monday.

Governors in other states put National Guard troops on alert. "We're just asking people to be patient and be ready for a long haul. But we have a very aggressive power restoration program in place and I think we're ready," Virginia Governor Robert McDonnell told the CNN program "State of the Union."

In Washington, Obama said officials had assured him that they had all the resources they needed in place, and he stressed that "it is important for us to respond big and to respond fast" to the hurricane's onslaught.

"We're going to cut through red tape and we're not going to get bogged down in a lot of rules," said Obama, who was having to juggle both is re-election bid and his efforts to stay on top of the storm's impact just nine days before Election Day.

Sandy blew the presidential race off course, forcing Obama and Republican challenger Mitt Romney to cancel scheduled campaign stops. It fueled fears that the storm could disrupt early voting ahead of the November 6 election.

INSURERS PREPARE FOR STORM

U.S. stock exchanges and Wall Street banks were sending employees into Manhattan on Sunday to stay in hotels and co-workers' homes, as markets prepared to open for business on Monday even as Sandy brought public transportation to a halt.

Insurers also prepared for the storm's arrival, activating claims teams, staging adjusters near the locations most likely to be affected and generally getting ready to pay for a potentially huge volume of losses.

While Sandy's 75 mph winds were not overwhelming for a hurricane, its exceptional width means the winds will last as long as two days, wearing down trees, roofs and buildings and piling up rainfall and storm surge.

Hurricane-force winds extended 175 miles from the center of the asymmetrical storm, while its lesser tropical storm-force winds spanned 850 miles in diameter.

"That's gigantic," said Chris Landsea, the hurricane center's science and operations officer.

At high tide, it could bring a surge of seawater up to 11 feet above ground level to Long Island Sound and New York Harbor, forecasters said.

"Given the large wind field associated with Sandy, elevated water levels could span multiple tide cycles, resulting in repeated and extended periods of coastal and bayside flooding," the forecasters said.

Sandy was centered about 250 miles southeast of Cape Hatteras, North Carolina, or 575 miles south of New York City at midday on Sunday, the hurricane center said. It pushed seawater up over the barrier islands off North Carolina known as the Outer Banks.

"It's flooded all over the village," longtime Ocracoke Island resident Kathleen O'Neal told Reuters. "I would say between a foot and two feet of water."

Sandy was moving over the Atlantic parallel to the U.S. coast at 10 mph, but was forecast to make a tight westward turn toward the U.S. coast on Sunday night.

Sandy killed at least 66 people as it made its way through the Caribbean islands, including 51 in Haiti, mostly from flash flooding and mudslides, according to authorities.

Job Description
Site Health Care Nurse Needed for Exelon Limerick Nuclear Plant , (Pottstown, PA)

PRIMARY PURPOSE OF POSITION
Plans, administers, provides, implements, and evaluates services to employees and contractors in company locations for clinical delivery, regulatory medical examinations and testing, drug and alcohol testing, immunizations, Fitness For Duty (FFD), pre-employment examinations, health counseling and teaching, disease prevention and wellness programs, OH & Services record documentation, activity and cost tracking reports and/or implements case management plans for workers compensation, urgent and emergency care services, short and long-term disability, and limited duty cases including rehabilitation and return to work plans. Responsible for after hour coordination of emergency and urgent care services delivery.

PRIMARY DUTIES AND ACCOUNTABILITIES
-Plans, schedules and provides medical testing and examinations including all appropriate resources, evaluates, records and communicates test results to meet regulation and policy requirements.
-Maintains confidential employee medical records, worker's compensation reports, activity and cost tracking reports using manual and computer based techniques.
-Analyzes data, identifies trends and develops programs to implement preventative and corrective measures addressing wellness and medical/work safety issues.
-Coordinates and communicates return to work transition plans with the case manager, and site and business unit personnel for occupational and non-occupational lost time and lost productivity issues.
-Performs urgent care and provides intervention for injuries and/or illnesses which impact the employee's or contractors ability to perform in the work place.

POSITION SPECIFICATIONS
Minimum:
-Registered Nurse.
-6 years Occupational Health Nursing
-Knowledge and understanding of the provision of case management for workers compensation, disability, limited duty, safety, industrial hygiene, healthcare plans, regulatory medical requirements and external healthcare resources, and regulation interpretation.
-Physical assessment skills and knowledge and experience in evaluation of medical screening test data.
-PC skills
-Certified in Spirotomy.
-Certified in Hearing Conservation.
-Certified Occupational Health Nurse.
Preferred:
-Bachelor of Science Degree.
-Site Clinical Experience in Nuclear Industry.
-Certified Case Manager.

2012 NFL Power Rankings, Week 7

Greetings, readers. ?Week 7 has come and gone and it feels like not much of interest happened. ?Sure, there was a bit of controversy in Tampa (due to an odd rule about going out-of-bounds and coming back in) and some OT drama, but the matchups really were not prime time stuff. ?Let's get to the stories, though.

As usual, we're starting with the Thursday night game. ?As usual, it wasn't terribly memorable. ?This week, it was more due to the playing style of the two teams involved. ?It angers me to watch a game where running the ball and defense is prioritized over throwing the ball. ?The NFL is a passing league. ?Take advantage of the rules that Goodell has put in! ?The commissioner and the rules committee has made it so that throwing the ball is very easy. ?And yet you have teams like Seattle and San Francisco either running the ball all day or dinking-and-dunking; either way, it translates to boring football. ?And the game on Thursday was a defensive struggle where the offenses looked mostly inept. ?That is boring to me. ?Oh, and by the way, the 49ers won.

Most disappointing game of the week? ?Baltimore at Houston, no contest. ?The Ravens didn't show up until the second half, and even then they weren't all that impressive. ?When the safety happened in the 1st quarter, I had a bad feeling, but I thought Flacco could maybe get a breather at halftime and come out ready for a comeback. ?Oops! ?So, once again, we have a guy who thinks he's elite and isn't. ?I remain unconvinced about the Texans, though, because they didn't show up the week before against Green Bay (also at home, it should be noted). ?But it is Baltimore that has really slipped in my estimation.

Minnesota and the NY Giants both won their games and now they're 5-2. ?Given Christian Ponder's recent play, it's hard to say that the Vikings are for real; they're another team that relies on the run for the most part. ?However, where they get an exemption from most people (and I suppose I'm included in that group) is that they have an All-World guy at RB. ?Adrian Peterson is a beast. ?The Giants pulled out a close one against Washington. ?I was impressed by the play that won the game for the Giants; Eli Manning lofted one to Victor Cruz and the salsa dancer did the rest, running it in for a TD. ?Much as I dislike the Giants (especially Eli), they played well and beat a pretty good Washington team. ?Well, Washington's offense is pretty good. ?Their defense is pretty stinky.

There were a couple OT games this week as well. ?New England went to OT against the Jets (at home, no less) while Oakland beat the Jaguars in OT. ?The Patriots blew a lead and looked bad down the stretch, but were able to rally in OT. ?Oakland turned a recovered fumble into the game-winning FG. ?Good to have a win, but neither game should have needed OT, really.

Finally we have the Chicago Bears. ?Despite getting knocked out for a play and then leaving the field before halftime, Jay Cutler seemingly willed the Bears to victory even while struggling offensively. ?He also looked pretty tough. ?That's not something I normally say about a guy I consider to be one of the biggest whiners in the NFL, but I try to be fair. ?Chicago is now 5-1 and look like the team to beat in the NFC North. ?I'm not entirely sold on them, though; while their defense is solid, I've never been a fan of Cutler and think that he's just a few INT's away from letting this season fall into the toilet. ?But right now, they're looking good.

?Well-rested and heading to Philly. ?7-0.

?They've been on both sides of a beatdown and now get a bye.

?Woeful Carolina comes to town. ?Bears shall feast.

4.) NY Giants (5-2) [+1]
?Giants head to Dallas to attempt to beat the team that got them in Week 1.

5.) San Francisco (5-2) [+1]
?A big Monday night game against Arizona could help determine the division winner.

6.) Minnesota (5-2) [+2]
?The Vikings get their crack at a Thursday night game. ?Let's hope they fare better than Seattle.

7.) Baltimore (5-2) [-5]
?A bye after a stunning beatdown. ?I guess that's a good thing, but too much stewing may hurt more than help.

8.) Green Bay (4-3) [+2]
?An easy game with Jacksonville coming in. ?Three in a row for Rodgers &co.

9.) Arizona (4-3) [-2]
?Falling off the pace dramatically, Arizona needs a win to "right the ship".

10.) Seattle (4-3) [-2]
?Falling off the pace dramatically, but has a better chance of getting a win.

Ill. Catholic Charities seek new path without foster care, adoption

The Catholic Charities agencies of Illinois are trying to meet society?s needs ?in a different way? in their first year without the state contracts that supported their adoption and foster care services.

?It?s been an interesting year,? Robert Gilligan, Executive Director of the Catholic Conference of Illinois, told CNA Oct. 17. He said agencies are ?continuing to adapt very well.?

The agencies are ?smaller? but are ?continuing to try to meet the needs of the most poor and vulnerable in our midst.?

Last year Illinois Catholic Charities were forced to close their adoption and foster care programs.

In July 2011 the Illinois Department of Children and Family Services ended the contracts under the state?s new civil unions bill. The department said the agencies? practice of placing children only with married couples discriminated against unmarried and homosexual couples. Lawsuits and legislative efforts attempting to preserve the decades-old collaboration between the Catholic agencies and the government failed.

The state contracts with the Catholic agencies totaled over $30 million annually and helped care for about 2,000 foster children.

The contracts? end had major consequences.

Catholic Social Services, the Catholic Charities agency in the Diocese of Belleville, split from the diocese to become Christian Social Services. It now accepts couples in civil unions as foster parents.

Catholic Charities of Peoria, which received $14.8 million of its $24 million budget from the state, went from nearly 400 employees to fewer than 20. Most of its employees left to work for the Center for Youth and Family Solutions, which took over the foster care cases of nearly 1,000 children.

Peoria?s Catholic Charities is restructuring to operate without any funding from the state. [More]

Markets on defensive amid US unease

LONDON (AP) ? Stocks suffered one of their worst sessions in weeks on Tuesday following another batch of weak U.S. corporate earnings and uncertainty over the outcome of the race for the White House.

So far, quarterly earnings coming out of the U.S. have been mixed, particularly from the technology sector. Downbeat updates on Tuesday from chemical company DuPont and manufacturer 3M added to the prevailing uncertainty.

"We are currently in the third week of a disappointing corporate earnings season," said Craig Erlam, market analyst at Alpari.

Facebook unveils its quarterly earnings after the markets close in a statement that could be crucial for the social network's future. The company has had a dire time since its stock flotation in May, losing more than a third of its value as investors became concerned about the company's ability to raise money from advertising.

"The results have the potential to make or break Facebook as an investment," said Erlam.

Over-arching the focus on earnings is the battle for the White House, with opinion polls showing the two candidates are in a dead heat. Last night's debate between President Barack Obama and Mitt Romney appears to have done little to change that.

"The accompanying uncertainty of being able to make any early calls over a likely winner is certainly going to create an air of unease across the board," said Fawad Razaqzada, market strategist at GFT Markets.

By mid-afternoon in Europe, Germany's DAX was down 2 percent at 7,185 while the CAC-40 in France fell 2.1 percent to 3,412. The FTSE 100 index of leading British shares was down 1.3 percent at 5,806.

In the U.S., the main indexes were close to seven week lows with the Dow Jones industrial average 1.8 percent lower at 13,106 while the broader S&P 500 index was off 1.7 percent to 1,409.

In recent weeks, markets have been largely buoyant amid signs that Europe was getting a grip on its debt crisis. Hopes that Spain will soon tap a new bond-buying facility from the European Central Bank have helped calm jitters over the eurozone's fourth largest economy but so far the Spanish government has made no request.

With its economy in a deep recession ? figures from the Spanish central bank showed the country's economy shrinking by a further quarterly rate of 0.4 percent in the third quarter ? investors are getting fidgety again. The yield on the country's 10-year bonds, a gauge of investor unease, has risen for the second day running, up a further 0.10 percentage point to 5.58 percent.

The euro has also taken a pounding, falling 0.8 percent at $1.2968, its first move below $1.30 since Oct. 15.

Earlier in Asia, stock indexes fared slightly better. Japan's Nikkei 225 index eked out a marginal gain to close at 9,014.25 a day after the country's currency fell to a three-month low against the dollar.

A weak yen helps Japan's mammoth export sector by raising the value of company profits repatriated from abroad. Still, grim export data ? which Monday showed a 10 percent drop in exports for September compared with a year earlier ? kept advances in check.

South Korea's Kospi declined 0.8 percent to 1,926.81 but mainland Chinese stocks lost ground, with the Shanghai Composite Index down 0.9 percent to 2,114.45. The Shenzhen Composite Index lost 1.4 percent to 869.96.

Hong Kong markets were closed for a public holiday.

Oil prices took a hit amid the increasing market gloom ? benchmark oil for December delivery was down $2.39 to $86.21 per barrel in electronic trading on the New York Mercantile Exchange.

No Twitter record, #bindersfullofwomen: Tuesday's debate in social

The first presidential debate set a new record for Twitter traffic around a U.S. political event--so expectations were high that Tuesday's town hall debate might set a new mark. But at the end of the debate, the official tally from Twitter was a mere 7.3 million tweets--around 3 million shy of the record--sent over the course of the evening. The moment that saw the most discussion was the exchange between the candidates over immigration.

Our folks at The Signal took a look at the number of mentions each candidate received. Former Gov. Mitt Romney had the most mentions, but President Barack Obama surpassed the number of mentions he garnered at the first debate, while Romney declined slightly.

Romney's comment about "binders full of women" during his discussion about attempting to secure gender equality in his cabinet while governor of Massachusetts was an immediate hit with the internet crowd--it was trending on Twitter in the United States shortly after he made the remark. Why settle for a binder full of women, BuzzFeed executive editor Doree Shafir quipped, when you could have a Trapper Keeper? Or, added Elizabeth Spiridakis, a retro FiloFax?

As for Romney's advisors take on the moment: According to Grace Wyler of Business Insider, even senior campaign advisor Eric Fehrnstrom had trouble suppressing a laugh when discussing it after the debate in the spin room.

The binder soon had a Tumblr and a Facebook page devoted to it. Within an hour after the debate's end, the Facebook page had over 90,000 likes. Thanks to Romney's choice phrasing, Patrick Swayze (OK, 1980s Swayze) was given an opportunity to weigh in on the 2012 fight for the White House.

To the joy of Newcastle fans around the world, the club have begun the process of tearing down the Sports Direct signs to return the club's historic name: St James' Park.

There was outrage when the club announced a sponsorship deal with the sportswear retailer which included the naming rights to the stadium to end 119-years of history in the city.

And anger directed at Mike Ashley - owner of both club and retail giant - in a move the fans regarded as running roughshod over the iconic heart of the city.? ?

Newcastle fans were quick to express their delight at the news on Twitter.

One Magpies supporter, ?@JeromeWalcott, said: 'Thank God St James Park has reverted back to it's rightful name. Should never have happened in the first place.'

And @SJ_NUFC simply said: 'The beauty that is St James' Park, is once more! #NUFC.'

Controversial new sponsors Wonga.com have recently signed a ?32million four-year shirt and stadium sponsorship deal.

The deal does not start until next season, but the club have immediately pulled down the Sports Direct signs and restore the St James? Park name.

The original lettering is due to be restored for Newcastle?s next home game, in the Europa League against Club Brugge. The move is a victory for supporters who have refused to acknowledge the change to the Sports Direct Arena.

However, Toon army are furious with plans to have the short-term loans company's name emblazoned on the front of the famous black and white stripes.

And one local MP has threatened to boycott the Wonga Arena if Ashley renames the stadium for a second time as part of a three-year deal worth a reported ?8m a year.

Travel Approval!


We are China bound! ?October 3rd brought the long awaited email with our official Travel Approval... the notice telling us we can go pick up our little boy.

Before we could purchase tickets we needed an appointment at the US consulate in Guangzhou. ?October 10th we finally got our confirmed date... November 6th!

Yesterday was a busy day communicating with our travel agent about flights, milage plus seats available on certain flights, layovers, stopovers and Jack Nathan's ONE WAY ticket. ?By 5pm we had secured our spots on flights to China and home again. ?We leave on October 23rd early in the morning!

This adventure has had so many blessings that we didn't expect or even think about when we first sent in our adoption application. ?The Lord has worked out every detail to get us to this point! ?Family and friends have rallied around our family and provided support of all types. ?Prayer, financial gifts, fundraising support, baby shower and milage plus tickets!! ?We are overwhelmed by love.

As we pack and prepare our family for this trip, our hearts are heavy for little Jack. ?His world is about to be turned upside down. ?Leaving his first home and all the people he loves will be very, very tough. ?Although we don't really know what to expect, we've been busy reading and learning as much as we can about toddler adoption and attachment resources.

Ultimately we trust that our Heavenly Father has brought us together and He will make us a family. ?It's going to be a tough road, but we are as ready as we'll ever be! ?And can't wait to hold our sweet little boy!!

Homemade Pasta | All Food, All Drinks, All Love

So, I finally did it: made my own pasta. I?m officially? just another person that made their own pasta! Maybe it?s just me, but making homemade pasta just seems a little more extraordinary than some other culinary creations. Just me? Okay, never mind. I enjoyed it, though. Plus, it?s really quite simple. Then again, I just love working with flour and dough!

Here I?ve got my four dough pieces rolled out into rectangles- well, they?re supposed to be anyway, but mine look more oblong than anything. Oh, it doesn?t matter? they taste just the same! (I even tasted a tiny tad of dough at this point? I had to make sure it was tasting good for the end product.) Then, just cut each one into strips about a quarter inch wide and hang ?em up like this.

Clothes hangers worked beautifully. Only, my pasta had broken at the top where it overhangs and fallen to the counter while drying overnight. I really didn?t care because it?s so much easier to eat shorter pasta anyway! But, I couldn?t quite explain why. Oh well. Time to cook it- and it was yummy! Good thing there was some leftover spaghetti meat to toss on top of my pasta because I wanted to sort of test it before preparing a whole planned-out meal for it. You understand? Exactly. So, now I?m ready to make more homemade pasta. It?s fun, it?s tasty and it?s totally awesome. Whoo!

Recipe:

2 c. flour

1 ts. dried basil or 1 ts. marjoram or 1 ts. sage

1/2 ts. salt

2 beaten eggs

1/3 c. water

1 ts. cooking oil or olive oil

Mix dry ingredients in a large bowl. In a small bowl, whisk wet ingredients. Make well in dry mixture, pour in wet stuff and mix well. Generously flour counter and dump sticky dough onto it, then add a little more flour on top of the dough. Knead till smooth and elastic, about 8-10 minutes. Let rest 10 minutes, covered. Divide into four equal parts. Roll each out to about 1/16 inch thickness. Let rest about 20 minutes, uncovered. Cut into desired pasta. Lie on wire rack or hang to dry. Store chilled for up to three days if dried completely. Or, dry an hour, then freeze. Cook dried or frozen pasta in boiling water with a pinch of salt for about 7 minutes, or until al dente. Use in your favorite pasta dishes!

Animals' microbial communities linked to their behavior

ScienceDaily (Oct. 11, 2012) ? New research is revealing surprising connections between animal microbiomes -- the communities of microbes that live inside animals' bodies -- and animal behavior, according to a paper by University of Georgia ecologist Vanessa O. Ezenwa and her colleagues. The article, just published in the Perspectives section of the journal Science, reviews recent developments in this emerging research area and offers questions for future investigation.

The paper grew out of a National Science Foundation-sponsored workshop on new ways to approach the study of animal behavior. Ezenwa, an associate professor in the UGA Odum School of Ecology and College of Veterinary Medicine department of infectious diseases, and her coauthors were interested in the relationship between animal behavior and beneficial microbes.

Most research on the interactions between microbes and their animal hosts has focused on pathogens, Ezenwa said. Less is known about beneficial microbes or animal microbiomes, but several recent studies have begun to explore these connections.

"We know that animal behavior plays a critical role in establishing microbiomes," she said. "Once they're established, the microbiomes then influence animal behavior in lots of ways that have far-reaching consequences. That's what we were trying to highlight in this article."

Bumble bees, for example, obtain the microbes they need through social contact with nest mates, including consuming their nest mates' feces-a not uncommon method for animals to acquire microbes. Green iguanas establish their intestinal microbiomes by feeding first on soil and later on the feces of adult iguanas.

"There are a lot of behaviors that animals might have that allow them to get the different microbes they need at different points of their lives," Ezenwa said.

Microbes, in their turn, influence a wide range of animal behaviors, including feeding, mating and predator-prey interactions.

One recent study found that fruit flies prefer to mate with others that have microbiomes most similar to their own. Another found that African malaria mosquitoes were less attracted to humans who had a greater diversity of microbes on their skin, possibly because certain microbes produce chemicals that repel these mosquitoes.

Other studies have focused on understanding the mechanisms by which microbes influence behavior.

"Recent experiments have been able to assess the molecules that are involved in communication between microbes in the gut and the brain of mice, showing that microbes are associated with shifts in things like depression and anxiety in these mice," she said. "There are huge implications in the role these microbes play in regulating neural function."

Ezenwa's own work involves investigating how social behavior and interactions between organisms might increase their likelihood of acquiring parasites and pathogens. She is starting a new project examining animal behavior and microbiomes in relation to infectious disease.

"As in the example of the bumble bees, behavior might control the microbes an animal acquires, and those microbes might then influence the animal's vulnerability to pathogens," she said.

The authors conclude that it will take a combination of molecular and experimental approaches to answer questions about the complex interactions between microbiomes and animal behaviors.

"This is a new, emerging topic that's worthy of much more investigation," Ezenwa said.

The article's coauthors are Nicole M. Gerardo of Emory University; David W. Inouye of the Rocky Mountain Biological Laboratory and the University of Maryland; Monica Medina of the University of California, Merced; and Joao B. Xavier of the Memorial Sloan-Kettering Cancer Center.
